Second, I have a Highscreen Realscan flatbed scanner. By digging in the
archives, I get the impression that this scanner is can be utilized by
the Microtek-driver.
...
- I cannot acquire preview in Color mode. It shows three smaller images
side by side, which somehow looks like the image ...
...

You are not using the current version of SANE/Microtek backend.
Get SANE-0.72 (which has v0.6 of the backend) and see what happens.
(My guess is that you have version 0.4 or 0.5 of the backend.)

The "RealScan" is really a ScanMaker E2 --- which for some reason declares
itself to have 1-pass capabilities, while it is a 3-pass scanner. Current
versions of the backend (starting with v0.6) fix this.

If it still doesn't work with the newer software, send a debug log (follow
the directions on the sane-microtek man-page), and indicate which versions
of the software, OS, scsi card, etc, you have (it can make a difference).
